BCMA (B-cell maturation antigen)/ TNFRSF17 (tumor necrosis factor receptor superfamily member 17) is a member of the TNF-receptor superfamily and are expressed preferentially expressed in mature B lymphocytes and may be important for B cell development and autoimmune response. They are also expressed on the surface of cancer cells. The Human BCMA/TNFRSF17 Protein is specifically designed to mimic these receptors, allowing it to be used in CAR T-cell therapy research

The CAR consists of an extracellular domain that recognizes the BCMA/TNFRSF17 protein, a transmembrane domain, and an intracellular domain that activates the T-cell when engaged by the target antigen. The Human BCMA/TNFRSF17 Protein is employed to evaluate the effectiveness of CAR T-cells in targeting and killing cancer cells expressing BCMA/TNFRSF17.

The addition of FC enhances the stability and solubility of the protein while incorporating Alexa Fluor 647, allows for a photostable antibody for precise visualization and detection of the protein in CAR T-cell assays.

Molecular Mass  The recombinant human TNFRSF17 consists of 303 amino acids and predicts a molecular mass of 33.3 kDa. In SDS-PAGE under reducing conditions, the apparent molecular mass of human TNFRSF17 is approximately 33-44 kDa due to the glycosylation.

Preparation and Storage

Shipping  The product is shipped at 2-8°C. 
Stability & Storage  To provide optimal stability of reagent:  

  • Store undiluted at 2-8°C 
  • Avoid prolonged exposure to light 
  • Avoid repeated freeze-thaw cycles. 


FACS Protocol

  • Reagents Required
    • Phosphate Buffered Saline (PBS with a pH ~7.4). 
    • FACS Buffer (PBS containing 2% of BSA). 
    • Human BCMA / TNFRSF17 Protein, Fc Tag, Alexa Fluor 647
  • Preparation of Reagents
    1. Human BCMA / TNFRSF17 Protein, Fc Tag, Alexa Fluor 647
      • Protein must be diluted before use for FACS.
      • Dilute the protein at a dilution of 1:100 in FACS buffer. 
  • Staining Protocol
    1. Harvest the cells from source and wash the cells once using FACS buffer. 
    2. Perform a cell count and a cell viability count. Cell viability must be ≥ 95% with a cell number above 2×105 live cells.
    3. In a round-bottom test tube, resuspend cells in 100 µL of diluted Human BCMA / TNFRSF17 Protein, Fc Tag, Alexa Fluor 647 for 30 min at 4°C.
    4. Wash the mixture of cells and antibody 3 times by centrifuge using FACS buffer.
    5. Resuspend the stained cells in 200 µL PBS per cell sample.  
    6. Transfer the stained cell into a desired flow tube for analyzation on desired Flow Cytometer. The recommended acquisition rate is >10,000 events.
